Protective Role of Vitamin C against the Histopathological and Ultrastructural Changes Induced By Imidacloprid in Testis of Male Rats

Abstract—In the present study, the effect of oral administration of imidacloprid over 4 weeks on histological and ultrastructural alterations in testis was assessed. Forty eight male rats were divided into four groups of 12 animals each. Group 1 was control, group 2 was administered with imidacloprid at the rate of 80 mg/kg b.wt/day, group 3 was fed with vitamin C at the rate of 10 mg/kg b.wt/day and group 4 was treated with both imidacloprid and vitamin C for 4 weeks. Histopathology of testis in group 2 revealed vacuolation, degeneration of seminiferous tubules, germinal cells were detached from basement membrane, increased interstitial spaces, disrupted basement membrane, presence of very few leydig cells, severe congestion in interstitial spaces and tunica albuginea was observed. Ultra thin sections of testis in imidacloprid treated group showed swollen nucleus, increased perinuclear space, varied size and shape of mitochondria and degeneration of spermatids. Testis of group 4 revealed swollen nuclei, margination of chromatin material, altered shape and size of mitochondria, degeneration of spermatids. In summary, these results suggest that exposure to imidacloprid (80 mg/kg) in male rats induced histological and ultrastructural alterations in testis and co-administration of vitamin C brought moderate protection.
